Khanpur lies in Rahim Yar Khan District in the south of Pakistan's Punjab province, on the main railway line between Lahore and Karachi. The railway has long been central to the town's growth, and on the map it cuts a long, straight line through the built-up area, with roads drawn toward the station from both sides.
The surrounding countryside is intensively farmed, watered by canals drawn from the Indus river system. Cotton, sugarcane and wheat are grown widely across the district, and the fields around Khanpur create a regular geometry of plots and channels. To the southeast lies the Cholistan desert, where irrigated land eventually gives way to sand.
